Phytosphingosine biosynthesis differs from sphingosine
in fish leukocytes and involves a transfer of methyl groups
from [3Hmethyl]methionine precursor. Biochem Biophys Res
Commun 250: 88-93. The incorporation of radioactivity
from either [3- 3H]serine as the direct or [3H-methyl]methionine
as the indirect precursor into sphingoid bases of free ceramides
in lymphocytes from fish was studied. Radioactivity from serine
was incorporated mostly in the sphingosine moiety of ceramides.
In contrast, the radioactivity from methionine was exclusively
incorporated into phytosphingosine base (i.e., 4-hydroxy-sphinganine)
and the incorporation increased by about twofold in the presence
of folic acid or niacinamide. |

Biochim Biophys Acta 1438(3):30521. Knowledge of sphingolipid
metabolism and function in Saccharomyces cerevisiae
is growing rapidly. Recent evidence suggesting that exogenous
sphingoid longchain bases must first be phosphorylated and
then dephosphorylated before incorporation into ceramide.
Phenotypes of strains defective in sphingolipid metabolism
are discussed because they provide hints about the undiscovered
functions of sphingolipidsand are one of the major reasons
for studying this model eukaryote. The longchain base phosphates,
dihydrosphingosine1phosphate and phytosphingosine-1phosphate,
have been hypothesized to play roles in heat stress resistance,
perhaps acting as signaling molecules. Dickson and Lester
evaluate the data supporting this hypothesis and suggest future
experiments needed to verify it. Finally, Dickson and Lester
discuss recent clues that may help to reveal how sphingolipid
synthesis and total cellular sphingolipid content are regulated. |
